摘    要:目的建立HPLC同时测定芫花药材中伞形花内酯、芫花素-5-O-β-D-茜黄樱草糖苷、芫花素-5-O-β-D-吡喃葡萄糖苷、木犀草素、椴苷、芹菜素、羟基芫花素及芫花素含量的方法。方法色谱柱:Diamonsil C18(250 mm×4.6 mm5,μm),流动相:甲醇(A)-体积分数为0.8%的醋酸水溶液(B),梯度洗脱(0~15 min:45%A、15~20 min:45%A~51%A、20~40 min:51%A、40~42 min:51%A~82%A4、2~60 min:82%A),流速:0.8 mL·min-1,波长:338 nm。结果伞形花内酯、芫花素-5-O-β-D-茜黄樱草糖苷、芫花素-5-O-β-D-吡喃葡萄糖苷、木犀草素、椴苷、芹菜素、羟基芫花素、芫花素的质量浓度依次分别在0.22~1.78、0.47~3.77、0.10~0.76、0.22~1.72、0.49~3.94、0.86~6.84、0.88~7.040、.44~3.56 mg·L-1内与峰面积呈良好的线性关系,平均回收率分别为98.7%、98.1%、97.4%、97.7%、101.1%、101.7%、100.3%、98.7%、RSD分别为2.7%、1.0%、2.4%、1.3%1、.8%、1.2%、1.6%2、.2%(n=5)。结论该法可为芫花药材的质量控制和临床用药安全提供依据。

Abstract:Objective To develop a method for the simultaneous quantitative analysis of umbelliferone, genkwanin-5-O-β-D-primeveroside, genkwanin-5-O-β-D-glucopyranosid, luteolin, tilirodide, apigenin, hydroxygenkwanin and genkwanin of Flos Genkwa. Methods Chromatographic separation of Eight compounds was achieved on a DIAMONSIL-C18 column(250 mm×4.6 mm, 5 μm).The mobile phase consisted of methanol (A) and 0. 8% HAc(B), gradient elution(0-15 min,45% A;15-20 min,45% A-51% A;20-40 min,51% A;40-42 min,51% A-82% A;42-60 min,82% A).The flow rate was 0.8 mL·min-1 and the detector wavelength was set at 338 nm.The column temperature was ambient temperature. Results The linear ranges of umbelliferone, genkwanin-5-O-β-D-primeveroside,genkwanin-5-O-β-D-glucopyranosid, luteolin, tilirodide, apigenin, hydroxygenkwanin and genkwanin were 0.22-1.78 , 0.47-3.77, 0.10-0.76 ,0.22-1.72 ,0.49-3.94 ,0.86-6.84 ,0.88-7.04,0.44-3.56 mg·L-1, respectively. The average recoveries (n=5) of the eight compounds were 98.7% with RSD of 2.7%,98.1% with RSD of 1.0%,97.4% with RSD of 2.4%,97.7% with RSD of 1.3%,101.1% with RSD of 1.8%,101.7% with RSD of 1.2%,100.3% with RSD of 1.6%,98.7% with RSD of 2.2%, respectively. Conclusion This method can serve as a useful quality control tool for Genkwa Flos .
