骨肉瘤中IGF-2基因印迹状态与临床关系的研究

摘    要:目的探讨骨肉瘤组织中,人胰岛素样生长因子-2(insulin—likegrowth factors2,IGF-2)基因的印迹状态及其与各临床病理因素间的关系。方法应用聚合酶链反应、逆转录PCR及限制性内切酶酶切等方法,检测30例骨肉瘤组织中IGF-2基因的印迹状态,分析IGF-2基因印迹丧失(loss of imprinting,LOI)及其与骨肉瘤各临床病理因素之间的关系。结果骨肉瘤组织中IGF-2基因印迹的丧失率为48.5%,瘤近旁组织中IGF-2基因印迹丧失率为21.2%,瘤远旁组织中IGF-2基因印迹丧失率为12.1%。同时,在有肺转移的骨肉瘤病例中IGF-2基因印迹丧失率(60.9%)明显高于无肺转移者(20%,P〈0.05)。结论IGF-2基因印迹丧失是贯穿于骨肉瘤发生、发展全过程的表观遗传异常,其与骨肉瘤的临床及患者预后相关。

Abstract:Objective To investigate the IGF-2 imprinting status in tissues of Osteosarcoma patients and its relationship with the other clinical/pathological parameters in Osteosarcoma. Methods IGF-2 imprinting status was determined by PCR and RT-PCR,followed by restrictive endonuclease (Apa I) digestion in 30 patients with Osteosarco ma. The loss of imprinting (LOI) of IGF-2,and its relationship with the other clinic opathologic parameters was studied in a group of prospectively recruited patients. Results LOI was positive in 5 of 11(48.5 %) of Osteosarcoma tis- sue;2 of 11(21.2% ) in adjacent neighboring tissue ;and 1of 11 (12.1% ) in distant neighboring tissue. The prevalence of IGF-2 LOI in cancer was significantly higher in patients with positive lung metastasis (60. 9%),as compare to those with negative lung metastasis (20.0%)(P〈0.05),respectively. Conclusion IGF-2 LOI has been confirmed to be associated with the initiation and development of Osteosarcoma ,which was related with clinical staging and prognosis.
