奥曲肽与胰岛素对人肝癌细胞影响的实验研究

摘    要:目的:探讨在体外胰岛素能否促进人肝癌细胞增殖,奥曲肽对其诱导的增殖活动有无抑制作用。方法:将奥曲肽与胰岛素直接作用于体外培养的人原发性肝癌细胞株,运用MTT、直接细胞计数及流仪测定等方法观察奥曲肽与胰岛素对肝癌细胞的活细菌数及其比值,细胞总数、细胞周期及增殖指数(PI)的影响。结果:胰岛素(0-5μg/ml)使人肝癌细胞株BEL0-7402的细胞总数、活细胞数及其比值增加;而奥曲肽(0-2μg/ml)则使细胞基础的及胰岛素刺激的细胞总数、活细胞数及其比值下降,5μg/ml的胰岛素增加细胞的PI值而1μg/ml的奥曲肽降低其PI值(P<0.05),并产生S期限滞作用。结论:在体外,胰岛素可诱导人肝癌细胞增殖,奥曲肽不仅直接抑制肝癌细胞的增,对胰岛素诱导的肝癌细胞增殖也有抑制作用。

Abstract:Objective To investigate in vitro whether the proliferative action of human hepatoma cell can be induced by insulin or inhibited by octreotide. Methods A human hepatoma cell line BEL-7402 was used. Using MTT method, cell count and flow cytometry (FCM), we observed the action of insulin (0-5 μg/ml) and/or octreotide (0-2 μg/ml) on the number of viable cells, its ratio, total number of cells, cell cycle and proliferative index (PI) in phase-synchronized cells.  Results The cell number, the viable cell number and its ratio in insulin (0-5 μg/ml)-treated BEL-7402 cells increased where as octreotide (0-2 μg/ml) decreased basic and insulin-induced ones in dose-dependent manner. One μg/ml of octreotide resulted in a reduced PI effect, which was increased in cells treated by 5 μg/ml of insulin. Moreover, the S cell cycle block effect was detectable in phase-synchronised cells subjected to treatment of 1 μg/ml octreotide. Conclusions In vitro, the growth proliferation of human heptoma cell can be induced by insulin, where as octreotide exerts inhibitory effect on basic and insulin-induced growth in human hepatoma cells.
