酵母蛋白质组研究进展

摘    要:近年来,数十种酵母基因组测序的完成,标志着酵母基因组研究的后基因时代的来临。而随着大量的DNA序列信息的获得,蛋白质组研究成为一种重要的研究酵母生物学机制的高通量方法。当前已经可以用二维凝胶电泳与质谱耦联的方法一次性研究大量的蛋白的表达,而高通量的酵母双杂交方法能够产生有价值的蛋白相互作用图谱。近来的蛋白微序列技术的发展进一步促进的蛋白相互作用的研究。本文主要介绍酵母蛋白质组研究的高通量技术手段及其在酵母中的应用。

Abstract:Recently, over a dozen yeast genomes have been sequenced, providing the basis to resolve several complex biological questions.Fueled by these ever-growing DNA sequence information, proteomics has become one of the most important disciplines for providing insight into the mechanisms of biological processes in a high-throughput mode.It is now possible to examine the expression of large amount of proteins using two-dimensional gel electrophoresis coupled with mass spectrometry technology.High-throughput yeast two-hybrid approaches have yielded valuable protein-protein interaction maps.Finally, recent developments in protein microarray technology provide a versatile tool to study protein-protein interactions.This review discusses high-throughput technologies for yeast proteome analysis and their applications.
