大鼠肾下腹主动脉缺血再灌注对心肌肌浆网钙泵活性及其基因表达的影响

摘    要:目的:观察大鼠肾下腹主动脉缺血再灌注对心肌肌浆网钙泵(sarcoplasmic reticulum Ca^2+ -ATPase,SERCA)的活性及其基因表达的影响。方法:复制大鼠肾下腹主动脉缺血再灌注(ischemia and reperfusion,I/R)模型。将SD雄性大鼠随机分为假手术组,缺血30min分别再灌注2、12、24和48h(I/R2h组、I/R12h组、I/R24h组、I/R48h组)。采用无机磷比色法检测缺血再灌注不同时间点心肌肌浆网SERCA活性,反转录酶PCR技术测定SERCA基因的表达。结果:I/R24h组、I/R48h组SERCA的活性及基因表达均冠著低于假手术组(P〈0.01);I/R12h组SERCA的活性均低于似手术组(P〈0.01),但基因表达无明显变化(P〉0.05);I/R2h组SERCA的活性及其基因表达与假手术组差异均无统计学意义(P〉0.05)。结论:肾下腹主动脉缺血再灌注过程严重影响SERCA的活性及其基因的表达。
