rhbFGF研制及促冠状动脉血管新生作用时间的研究

摘    要:目的 探讨重组人碱性成纤维细胞生长因子 (rhb FGF)促冠状动脉血管新生的作用时间。方法 开胸结扎兔冠状动脉左前降支 (L AD) ,建立急性心肌梗死动物模型 ;制备 rhb FGF蛋白 ,将其直接四点注入兔缺血心肌内 ,通过病理切片观察缺血心肌内血管新生情况。结果 结扎 L AD后 6周及 12周 ,rhb FGF组与生理盐水对照组 (NS)病理切片 ,随机观察 10个高倍视野无平滑肌小血管计数分别为 6周 (W) :41.13± 10 .0 4,30 .33± 3.2 1,P<0 .0 1;12 W:5 0 .5 0± 9.2 0 ,32 .6 3± 7.2 5 ,P<0 .0 1;有平滑肌血管计数分别为 6 W:16 .2 5± 5 .2 3,10 .75± 4.2 5 ,P<0 .0 1;12 W:16 .88± 4.87,11.2 5± 5 .0 9,P<0 .0 1。结论 缺血心肌内注射 rhb FGF可促进冠状动脉血管侧支新生 ,其作用时间可能小于 12 W。

Abstract:Objective To investigate the time of recombinant human basic fibroblast growth factor (rhbFGF) on promoting angiogenesis of coronary artery. Methods A rabbit model was made by opening chest and ligating the left anterior descending (LAD) coronary artery. rhbFGF was made by ourselves, and it was directly injected into ischemic myocardium with four points to observe the time and effects of angiogenesis by pathology histological sections. Results Six weeks after ligation, in rhbFGF and normal saline(NS) control groups the vascular counts with no smooth muscle cell were 41.13±10.04,30.33±3.21,P<0.01; 12 weeks:50.50±9.20,32.63±7.25,P<0.01, by 10 high-power fields of microscopy at random. Six weeks after ligation, the vascular counts with smooth muscle cell were 16.25±5.23,10.75±4.25,P<0.01; 12 weeks:16.88±4.87,11.25±5.09,P<0.01, respectively. Conclusions The effects of angiogenesis of rhbFGF by direct injecting into ischemic myocardium were related with the time, which maybe less than 12 weeks.
