益气活血法改善气虚血瘀证局灶性脑缺血再灌注模型鼠生物学特征的有效性评价

摘    要:目的:拟建立相对完备的症状、体征客观定量的评估体系,对益气活血法改善气虚血瘀证局灶性脑缺血再灌注模型鼠症状、体征的有效性方面作出客观评价。方法:采用饥饿、劳累、高脂饮食等多因素复合制作出气虚血瘀证模型,然后用线栓法阻断大脑中动脉制作出局灶性脑缺血再灌注模型。实验动物随机分为假手术组、模型组、益气组、活血组、益气活血组。在手术过程中对其存活率进行观察;分别在缺血再灌注6h、24h、3d对其神经系统体征进行评分;分别在缺血再灌注3d、7d对其体重、气虚、血瘀症状、体征做出客观评价;在缺血再灌注3d进行HE、免疫组化染色。结论:益气活血法能显著改善气虚血瘀证局灶性脑缺血再灌注模型鼠的生物学特性,抑制细胞凋亡可能是其临床疗效机制之一。

Abstract:Objective: It is intended to set up a relatively objective and quantiative system evaluating symptoms and bodily characteristics and to give objective evaluation of the validity of biological characteristics improvement of qi deficiency and blood stasis local cerebral ischemia reperfusion model mice with the treatment of yiqi huoxue. Methods: Qi deficiency and blood stasis model is made by means of combined factors of hunger, tiredness and high fat diet and afterwards local cerebral ishemia reperfusion model is completed by blocking middle cerebral artery with thread occluding ways. The test animals are divided into pseudo surgery group, model group, yiqi group, huoxue group and yiqi huoxue group. The mortality rate is observed;Neurological system symptoms is evaluated after 6h, 24h, 3d of ishemia reperfusion; Weight, qi 1 deficiency, blood stasis and physical characteristics after 3d, 7d reperfusion are objectively evaluated; HE and immunological staining is made after 3d of ishemia reperfusion. Results: The Yiqi huoxue treament may significantly improve the biological characteristics of local cerebral ischemia reperfusion model mice, and restraining cell apoptosis may be one of the clinical mechanism.
