摘    要:本文采用2,3-二氨基萘荧光法测定人尿(或鼠尿)中总硒含量。以往采用的HClO_4+H_2SO_4+Na_2MoO_4混合消化液消化尿样时,不能把正常及高硒地区尿中总硒全部转变成Se~(+4),特别是测定尿硒含量较高的样品,得出的结果明显偏低。本法改用HNO_3+HClO_4(+HCl)混合酸作消化液后,能全量地把尿中总硒转变成Se~(+4)。于尿样中加入不同量的TMSe~+-Se后,硒的回收率为95.9~103.4%。本法硒的最低检出量为2ng。人尿中硒含量在0.022~0.251μg/ml范围内时,测出结果的变异系数不超过5%。本方法适用于高硒尿中总硒含量的分析,结果稳定。

Abstract:Total selenium content in human urine was determined fluorometric-ally by 2, 3-diaminonaphthalene. It is found that the old digestive mixture HCIO_4+H_2SO_4+Na_2MoO_4 cannot completely convert urine selenium to Se~(+4) state in the urine samples from the normal and seleniferous areas. The results obtained were apparently low in urine samples from the seleniferous areas. In this method, HNO_3+HClO_4(+HlC)was used as a digestive mixture proposed by Nahapetian, it could quantitatively convert the urine selenium to Se~(+4) state. Coefficient of variation and recovery after adding TMSe~+-Se were less than 5% and 95.9-103.4% respectively. As low as 2ng selenium in the sample was detected. This method can be applied to the determination of total selenium in urine from the normal and high selenium areas.
