脑脉安胶囊对大鼠急性脑梗死损伤中BDNF mRNA表达的影响

摘    要:目的探讨脑脉安胶囊对大鼠急性脑梗死损伤大脑皮层中脑源性神经生长因子(BDNF)mRNA表达的影响。方法雄性SD大鼠36只,随机分为脑脉安胶囊低剂量组、脑脉安胶囊中剂量组、脑脉安胶囊高剂量组、西药组、假手术组、缺血24 h模型组共6组,每组6只。采用线栓法制作大鼠局灶性脑缺血模型,3只于24 h断头取脑,HE染色观察大鼠大脑皮层区细胞损伤程度,3只取出大鼠脑组织,提取脑组织中总RNA进行RT-PCR扩增BDNF mRNA特异性片段,分析各组BDNF mRNA的变化。结果缺血24 h模型组大鼠均出现明显的神经功能缺损症状,有明显缺血性形态学改变如细胞水肿等,BDNF mRNA扩增产物相对光密度较假手术组明显升高(P<0.01);脑脉安胶囊各剂量组及西药组皮层区神经细胞水肿减轻,BDNF mRNA扩增产物相对光密度均较缺血24 h模型组明显升高,具有统计学意义(P<0.01)。结论脑脉安胶囊通过上调脑源性神经因子BDNF,使受损的神经功能得到恢复,从而发挥其对治疗急性脑梗死的作用。

Abstract:Objective To investigate the effect of Naomaian Capsules on expression of brain-derived neurotrophic factor(BDNF) mRNA in acute cerebral infarction tissue of rats.Methods 36 SD male rats were randomly divided into 6 groups,with 6 rats in each group: low-dose Naomaian Capsules group(A),moderate-dose Naomaian Capsules group(B),high-dose Naomaian Capsules group(C),west medicine(nimodipine)group(D),sham-operation group(E),and 24h ischemia model group(F).The model of focal cerebral ischemia was made by thread-occluded method.In each group,the brain tissues of 3 rats were taken out 24 hours after the operation and the injuries of their cerebral cortex cells were observed by HE staining and the brain tissues of another 3 rats were taken out to extract the total mRNA for amplification of BDNF mRNA specific fragment by RT-PCR and analyze the changes of BDNF mRNA expression of each group.Results Obvious symptoms of neural function deficiency and ischemic morphological changes such as cellular edema were observed in 24h ischemia model group.Relative optical density of the amplified products of BDNF mRNA in 24h ischemia model group was obviously higher than in sham-operation group(P<0.01).The relief of cortex neuron edema and relative optical density of amplified products of BDNF mRNA in Naomaian Capsules group with all dosages and west medicine group were obviously higher than 24h is chemia model group,and the results had statistical significances(P<0.01).Conclusion Naomaian Capsules can up-regulate the expression of BDNF to cause the recovery of the neurological function,which contributes to its therapeutic effect for acute cerebral infarction.
