美洛西林与四种常用输液的配伍稳定性考察

摘    要:目的考察注射用美洛西林钠在4种临床常用输液中的稳定性。方法按临床常用剂量配制美洛西林钠的0.9%氯化钠注射液、10%葡萄糖注射液、5%葡萄糖注射液以及5%葡萄糖+0.9%氯化钠注射液,考察上述4种配伍液于0、0.5、1、2、3、4、5、6 h的pH值、颜色和澄明度外观变化。用高效液相色谱法考察在上述时间点下,配伍液中美洛西林钠的含量变化。采用BDS-C18(250 mm×4.6 mm,5μm)色谱柱,以磷酸盐缓冲液(pH值为5.8)∶甲醇(60∶40)为流动相,检测波长:210 nm,柱温:30℃,进样量:1μL。结果以本实验所建立的色谱条件对美洛西林钠进行检测,专属性好,能达到基线分离,美洛西林钠的线性范围为1~12 mg/mL(r=0.9999),日内精密度RSD<2.0%,4种输液配伍液在6 h内峰面积均无明显变化,变异系数RSD均<2.0%。结论美洛西林钠与0.9%氯化钠注射液、10%葡萄糖注射液、5%葡萄糖注射液以及5%葡萄糖+0.9%氯化钠注射液配伍后稳定,临床上可配伍使用。
