TNF-α蛋白和mRNA在大鼠药物性胃粘膜损伤表达的研究

目的：用小剂量阿司匹林灌服大鼠建立药物性胃粘膜损伤模型，探讨 TNF-α 蛋白和 mRNA 在胃粘膜细胞的表达及在胃粘膜损伤中的作用。方法：免疫组化和反转录聚合酶链反应技术检测 TNF-α 的表达。结果:实验组 TNF-α 在用药后 15 min 开始表达，3 h 达高峰，持续到 6 h，12 h 开始下降，24 h 有少量表达。实验组用药后与用药前比较 TNF-α 表达水平有明显差异。预处理组和对照组两组分别与实验组比较，TNF-α 均明显呈低水平表达。预处理组和对照组之间 TNF-α 表达水平无明显差异。结论:TNF-α 的表达在小剂量阿司匹林引起的胃粘膜损伤的发病机制中有重要意义。

Study on the expression of TNF-a protein and mRNA in the rat drug-induced gastric mucosal injury

Objective:To determine the expression of TNF-α protein and mRNA in rat gastric mucoasl cell during small dose aspirin-induced gastric mucosal injury and its signifieance. Methods: By immunohistochemistry and RT-PCR method, we examined the expression of TNF-α in gastric mucosal cell. Results: TNF-α started to express at 15min after administration of aspirin , peaked at 3h ,decreased at 12h, remained at 24h in experiment group. The expression of TNF-αdecreased significantly after administration of aspirin compaired with that before administration of aspirin in experiment group. There is lower level expression in control group and preced-dispoeed group than in experiment group. There is no obvious diffirence between control group and preced-disposed group Conclutions: The expression of TNF-α phys an important role in the mechanisms of small dose aspirin-induced gastric mucosal injury.
