鼻腔滴人IFN-γ对AR大鼠鼻腔黏膜中GATA-3表达的影响及意义

摘    要:目的 探讨鼻腔吸入IFN-γ对大鼠变应性鼻炎(AR)的治疗作用及其机制。方法将32只大鼠随机分为四组,各8只.A组为阴性对照组。B、C、D组采用卵清蛋白、氢氧化铝建立大鼠AR模型,B组为阳性对照组、C组为IFN-γ,治疗组、D组为丙酸倍氯米松治疗对照组。B、C、D组于模型建立后第31~38天,每只每日每侧鼻腔分别滴入PBS 50μ1、IFN-γ 1μg 和丙酸倍氯米松3.5μg。第39天取鼻腔黏膜,观察鼻腔组织病理学改变,用免疫组化SP法检测转录因子GATA3。结果与A组相比,B组鼻黏膜充血、水肿,黏膜层增厚,有嗜酸细胞为主的炎性细胞浸润,GATA3表达增加;C、D组上述炎性改变明显减轻,鼻黏膜中GATA3表达减少。结论鼻腔滴入IFN-γ治疗AR可能与阻断GATA3表达,从而继发抑制Th2型反应有关。

Treatment of allergic rhinitis rats by blockade of GATA3 expression with absorbed interferon gamma

Abstract:Objective To investigate the efficacy and mechanism of absorbed interferon gamma (IFN-γ) in treatment of allergic dfinitis(AR) in rats. Methods Ovalbumirl (OVA) and aluminum hydroxide were used to construct the AR model. 24 AR rats were randomly divided into positive control group( group B, n = 8 ), IFN-γ treatment group( group C, n = 8 ) and beclomethasone dipropionate treatment group ( group D, n = 8 ). The negative control group was consisted of 8 normal rats. After the AR models built,50 μl PBS, 1 μg IFN-γ and 3.5 and beclomethasone dipropionate was respectively dropped in each nasal cavity of every rat in group B,group C and group D from day 31 to 38 once daily. The nasal mucosa were collected on day 39 in order to observe the pathologic changes ,and the expression of GATA-3 were determined by immunohistochemistry SP. Results The nasal mucosa of AR rats was obviously of congestion, edema and thickening with inflammatory cells infiltration mainly of eosinophils in group B. But the above mentioned changes were much more ameliorated in group C and D. GATA3 expression was showed decreased in the nasal tissue in group C and D, significantly increased in group B. Conclusions The curative effect of IFN-γ might be involved the mechanisms of restraining the Th2 reaction by blockade of GATA3 expression in the nasal tissue in AR rats.
