摘    要:研究p53抑癌基因与大肠癌发生与发展的关系。应用原位分子杂交技术对62例大肠部区,癌移行及癌旁区大肠组织中p53癌基因进行检测。大肠癌区呈p53mNRA杂交阳性反应,阳性颗粒主要分布在癌细胞浆内,少部分阳性颗粒核浆共存,阳性癌细胞多呈灶状分布。

Abstract:Objective: To study the relation between the tumor suppressor gene p53 and the occurrence and development of colonic carcinoma. Methods: By use of in situ hybridization, the oncogene p53 ( p53 mRNA) was detected, in 62 cases of colonic carcinoma, in carcinomatous region, the transitional region and the normal colonic tissues surrounding carcinoma. Results: p53 mRNA was positive in the carcinomatous area. The positive granules were mainly intracytoplasmic in the carcinomatous cells. A small number of positive cells showed the coexistence of the nucleo-cytoplasmic granules. The positive carcinomatous cells were chiefly distributed in focuses. The positivity and positive intensity of p53 mRNA in carcinomatous region were higher than those in transitional region (P<0. 01), p53 mRNA was negative in the normal colonic tissues surrounding carcinoma. The positivity of p53 mRNA in the group with lymph nodes and liver metastasis was markedly higher than those in the group without metastasis. Conclusions: The results denote that the expression of p53 mRNA has a close relation to the occurrence and development of colonic carcinoma. The positive p53 mRNA in the carcinomatous could be regarded as biomolecular index for the evaluation of metastasis and prognosis in colonic carcinoma, and further as an appraisal of border decision in clinical practice.
