姜黄素对博莱霉素诱导大鼠肺纤维化的保护作用

摘    要:目的观察姜黄素对博莱霉素诱导大鼠肺纤维化的保护作用及其作用机制。方法 72只大鼠随机分为三组:正常对照组(气管内一次性注射生理盐水2 mL/kg)、博莱霉素组(气管内一次性注射博莱霉素5 mg/kg)和姜黄素组(气管内一次性注射博莱霉素5 mg/kg后第15天起每日腹腔注射姜黄素50 mg/kg)。分别在实验第14、17、21、28天每组各处死大鼠6只,取肺组织行苏木精-伊红(HE)和Masson染色以评价病理变化;双抗体夹心ELISA测定支气管肺泡灌洗液(BALF)中IL-10含量。结果姜黄素组第28天肺泡炎和肺纤维化程度显著轻于博莱霉素组(均P<0.05);BALF中IL-10含量显著高于博莱霉素组(P<0.05)。结论姜黄素可以减轻博莱霉素诱导的大鼠肺纤维化,其保护作用可能与促进IL-10表达有关。

Abstract:Objective To evaluate the protective effect and mechanism of curcumin against Bleomycin-induced pulmonary fibrosis in rats.Methods Seventy-two SD rats were randomly divided into three groups: the control group which received a single intratracheal dose of 2 mL/kg of sterile physiological saline;the Bleomycin group intratracheally instilled a single dose of 5 mg/kg of bleomycin;the curcumin group which received 50 mg/kg of curcumin intraperitoneally daily from the 15 days after Bleomycin administration.At 14,17,21,28 days,six rats per in treatment group were sacrificed.The sections of lungs were stained with hematoxylin-eosin(HE) and Masson′s trichrome to evaluate the pathological changes.IL-10 protein level in bronchoalveolar lavage fluid(BALF) was assayed by using a commercial enzyme-linked immunosorbent assay(ELISA) kit.Results The alveolitis and the pulmonary fibrosis were significantly alleviated in the curcumin group at 28 days(all P 0.05).Curcumin remarkably increased the concentration of IL-10 in BALF(P 0.05).Conclusion Curcumin can prevent bleomycin-induced pulmonary fibrosis,possibly by its increasing of IL-10 activity.
