Graves’病患者外周血T细胞表面膜型CD28及血清中可溶性CD28的表达及其意义

摘    要:目的分析Graves′病(Graves′disease,GD)患者外周血T细胞表面膜型CD28及血清中可溶性CD28的表达水平,探讨CD28分子在GD免疫病理机制中的作用。方法选取105例GD患者为试验组,67例健康者为对照组,通过溶血及流式细胞术检测所有受试对象外周血T细胞表面膜型CD28的表达,酶联免疫吸附法(ELISA)检测血清中可溶性CD28的含量。结果试验组外周血CD4+CD28+T细胞为(23.367±8.562)%,CD8+CD28+T细胞为(10.156±3.114)%,较对照组分别为(30.786±5.143)%和(17.200±3.624)%]显著减少(P<0.0.5、P<0.0.1),血清中可溶性CD28水平观察组(2.35±0.58)μg/L]较对照组(0.53±0.23)μg/L]显著增加(P<0.01)。结论共刺激分子CD28很有可能参与了GD的免疫病理机制。

Abstract:Objective To analyze the expression of membrane type CD28 on T cells and the serum production of soluble CD28 in peripheral blood from GD patients,to investigate the role of CD28 molecule in the immunological pathogenesis of GD.Methods The peripheral blood mononuclear cells(PBMCs) were collected from 105 cases of GD patients as experi ment group and from 67 healthy donors as control group.The expression of membrane type CD28 in peripheral blood of all donors was detected by flow cytometry(FCM).The production of soluble CD28 in serum was detected using the skills of enzyme-linked immunosorbent assay(ELISA).Results The percentage of CD4^+CD28^+ and CD8^+CD28^+ T cells in peripheral blood of patients from experiment group were respectively(23.367 ±8.562)% and(10.156 ±3.114)%,and were declined markedly compared with that of control group (30.786±5.143)% and(17.200±3.624)%,respectively](P〈0.0.5,P〈0.0.1).In addition,compared with control group (0.53±0.23)μg/L],the level of soluble CD28 was increased significantly in serum of experiment group (2.35±0.58)μg/L](P〈0.01).Conclusion The costimulatory molecule CD28 maybe take part in the immunological pathogenesis mechanism of GD.
