摘    要:目的 探索建立化疗所致卵巢早衰动物模型的方法。方法 以Wistar雌性大鼠为研究对象,以5种不同剂量和方案腹腔注射环磷酰胺,观察大鼠一般情况、动情周期、性激素(雌二醇和卵泡刺激素)水平、卵巢重量及形态学、各级卵泡数的变化。结果 环磷酰胺可引起大鼠体重明显下降,动情周期紊乱,低雌激素血症,高促性腺激素血症,卵巢重量明显减轻,卵巢间质增生明显,窦前及窦状卵泡数减少,与对照组相比均有明显差异。结论 采用腹腔注射环磷酰胺负荷剂量50 mg/kg后以8 mg/(kg ·d) 的维持剂量连续注射14d的方案建模,大鼠卵巢功能明显衰退,卵巢组织学及内分泌改变与临床卵巢早衰患者相似。该方法简便易行、成模时间短、成功率高、死亡率低,是建

Abstract:Abstract Objective To investigate an applicable method of establishment of animal model of chemotherapy-induce premature ovarian failure. Methods Female Wistar rats were injected i. p. with cyclophosphamide(CTX) at different doses and proposals. The body weight, estrous cyclicity, concentration of gonadal hormones(estrogen and follicle-stimulating hormone), ovarian weight and histopathology, number of follicles were observed, and compared with those of control group. Results cyclophosphamide induced decreased body and ovarian weight, abnormal estrous cyclicity, hypoestrogenemia, hypergonadotrophinemia, decreased preantral and antral follicles. Conclusion The proposal of load dose of CTX(50 mg/kg) followed by i.p. 8 mg•kg-1•d-1 for 14 days can damage the ovarian function. The changes of sex hormone and histopathology were similar to those of clinic premature ovarian failure patient. The method can be used to establish the animal model of chemotherapy-induced ovarian failure.
