Effect of Hypoxia on the Expression of Nodal and the Impact of Relative Migration Protein in Malignant Melanoma A375 Cells

Abstract:Objective To discuss the effect of hypoxia on the expression of Nodal and the impact of relative migration protein in malignant melanoma A375 cells.Methods Malignant melanoma A375 cells were cultured in vitro,the cells were divided into blank control group and CoCl2 group,SB-431542 group and CoCl2+SB-431542 group,the cells were treated with chloride(CoCl2)to construct an intracellular hypoxia model,SB-431542 blocks the Nodal signaling pathway,the level of Nodal,Activin receptor-like kinase 7(ALK7),matrix metalloproteinase-2(MMP2)and vimentin in malignant melanoma A375 cells in different environments was detected by Western blot.Results The cell morphology of A375 cells was not significantly changed after SB-431542 with different concentrations blocked the Nodal signaling pathway,there were statistically significant differences in the proliferation activity of A375 cells when the above concentrations were blocked for 24 h,48 h and 72 h(P<0.05),the cell activity increased significantly over time,but as the concentration increased,the cell activity decreased,the inhibition of SB-431542 on A375 cell proliferation was dose-dependent and time-dependent.The expression levels of Nodal,MMP2,ALK7 and Vimentin in each group were significantly different(P<0.05),the expression levels of Nodal,MMP2,ALK7 and Vimentin in A375 cells in CoCl2 group were all higher than those in blank control group(P<0.05),compared with CoCl2 group,SB-431542 group and CoCl2+SB-431542 group were decreased(P<0.05).Conclusion Hypoxia can increase the expression of Nodal,MMP2 and vimentin in the malignant melanoma A375 cells,it may be related to migration and invasion deterioration of A375 cells,At the same time,the Nodal signaling pathway of cell proliferation is blocked by SB-431542.
