Abstract:Objective:To analyze the expression of insulin-like growth factor-2(IGF-2)and insulin-like growth factor binding protein-3(IGFBP-3)in placenta of patients with fetal growth restriction(FGR),and explore their clinical significance.Methods:A total of 110 patients with FGR diagnosed by ultrasound and delivered in our hospital from December 2017 to December 2019 were selected as FGR group,and 110 normal full-term pregnant women with normal singletons who delivered at the same period were selected as the control group.The placental tissues of the two groups were collected.General data of patients were compared between the two groups.The expression levels of IGF-2 mRNA and IGFBP-3 mRNA in placental tissue were detected by qRT-PCR.The expression levels of IGF-2 and IGFBP-3 protein were detected by immunohistochemistry.Spearman correlation analysis was used to analyze the correlation between IGF-2 and IGFBP-3 proteins in placenta of patients with FGR,and the binary logistic regression was used to analyze the influencing factors of FGR.Results:In the comparison of the general data between two groups,the fetal weight and early pregnancy related plasma protein A(PAPP-A)in FGR group were significantly lower than the control group(P<0.05).Compared with the control group,the expression level of IGF-2 mRNA and positive expression rate of IGF-2 protein in the placenta tissue of the FGR group were significantly decreased(P<0.05),while the expression level of IGFBP-3 mRNA and positive expression rate of IGF-2 protein were significantly increased(P<0.05).Spearman correlation analysis showed that there was a negative correlation between IGF-2 and IGFBP-3 protein expression(P<0.05).The binary logistic regression analysis showed that the abnormally lower expression of PAPP-A and IGF-2 mRNA,and the abnormally higher expression of IGFBP-3 mRNA in placental tissues were related to the occurrence of FGR(P<0.05).Conclusions:The abnormally lower expression of IGF-2 protein and the abnormally higher expression of IGFBP-3 protein in the placenta tissue may have a certain impact on the pregnancy outcome,and which are related to the occurrence of FGR.
