Abstract:Objective:To study the inhibitory effect of PolyphyllinⅠon Huh7 and HepG2 hepatoma cell lines and to explore whether it has the inhibitory effect to hepatocellular carcinoma stem cells.Methods:After hepatoma cells were treated with different doses of PolyphyllinⅠ,MTT assay was used to detect cell survival,flow cytometry was used to detect cell apoptosis,Transwell was used to detect cell migration.CD133+-Huh7 and CD133+-HepG2 cells were separated by magnetic beads.The effect of Polyphyllin I on the sphere-forming ability of CD133+and CD133-hepatoma cells was detected by spheroidization test.Results:PolyphyllinⅠcould inhibit the proliferation and clone formation of Huh7 and HepG2 cells,and inhibit the migration of hepatocellular carcinoma cells.Moreover,PolyphyllinⅠcan induce dose-dependent apoptosis of hepatocellular carcinoma cells.The results of magnetic separation and pelletization in vitro showed that PolyphyllinⅠmade the cells smaller and the cell morphology shrunk,which had no difference in effect on CD133+-Huh7 and CD133+-HepG2,made cells were smaller,cell morphology shrivel.Conclusion:PolyphyllinⅠcan inhibit the proliferation,migration and induce apoptosis of Huh7 and HepG2 hepatocellular carcinoma cells,but it has no obvious the ability to kill hepatocellular carcinoma stem cells.
