ESN,AES在IGSS试验中作为dsDNA底物特异性的进一步研究

AFURTHER STUDY ON THE SPECIFICITIES OF ESN AND AES USED IN IGSS AS dsDNA SUBSTRATES

A further study on the specificities of enzyme-treated human spermatozoa nuclei (ESN) and acid-elutiou-treated substrates of mouse liver/kidney ( AES-liver/kidney ) as dsDNA substrates in immunogold-silver staining (IGSS) for anti-dsDNA antibodies ( AdsDNA ) was performed. It was found that the results of detection changed when the sera had been absorbed with DNA or when the substrates treated with deoxyribonuclease I. However, the results were not obviously changed when the sera had been absorbed with histone or RNA, or when the substrates heated or treated with trypsin, chymotrypsin or ribonuclease. These data suggest that it is dsDNA in the substrates that reacted to the relevant serum antibody ( AdsDNA).It was also found that the anti-spermatozoa positive serum and the serum from vasoligated man showed no positive reaction to ESN either . So the specificities of ESN and AES as dsDNA substrates in IGSS are definitely good.
