摘    要:目的研究经神经生长因子(NGF)基因修饰的骨髓间充质干细胞(BMSCs)对血管性痴呆(VaD)大鼠的作用。方法以NGF和绿色荧光蛋白(GFP)基因转染BMSCs;制备VaD模型;将大鼠随机分为假手术组、磷酸盐缓冲液(PBS)组及NGF修饰组。造模1周后,通过尾静脉注射将NGF基因修饰的BMSCs移植到大鼠体内;4周之后进行Morris水迷宫检测;以免疫组织化学法检测大鼠海马区NGF和SYN表达。结果与PBS组比较,NGF修饰组逃避潜伏期缩短显著,而SYN表达明显升高(P0.05)。结论经NGF基因修饰的BMSCs对VaD大鼠的学习和记忆能力有改善作用;并可以提高NGF及SYN的表达。

Abstract:Objective To investigate the effect of NGF gene modified bone marrow mesenchymal stem celts(BMSCs)transplantation into the brain of vascular dementia (VaD)rat. Methods The VaD models were established,and then they were randomly divided into 3 groups:sham group, PBS group and NGF group. Behavioral abnormalities were observed by Morris water maze (MWM) test. Then, the expression of synapto- physin and NGF was detected by the immunohistochemistry technique. Results Morris water maze test showed that the average escape latency in the NGF group was shorter than that in the PBS group, and the expression of synaptophysin and NGF was significantly higher than that of the PBS group(P〈0.05). Conclusion The transplantation of NGF gene modified BMSCs can improve the learning-memory ability and increase the synaptophysin and NGF expression in the VaD rat.
