人脑胶质瘤耐药基因mdr1、ERCC2、MGMT的表达及相关性分析

摘    要:目的:探讨人脑胶质瘤组织中三种耐药基因 mdr1、 ERCC2和 MGMT mRNA表达之间的相关性。方法:采用双管 RT- PCR方法检测 14例正常脑组织和 56例人脑胶质瘤组织中 mdr1、 ERCC2和 MGMT mRNA水平,并分析其相关性。结果:正常脑组织中 mdr1、 ERCC2和 MGMT的表达量分别为 (0.75± 0.36), (1.43± 0.92)和 (2.48± 1.83), mdr1和 ERCC2、 ERCC2和 MGMT之间具相关性 (P< 0.01和 P< 0.001);胶质瘤组织中三种基因表达量分别为 (0.53± 0.42), (1.75± 1.16)和 (2.88± 1.92), ERCC2和 MGMT之间具极显著相关性 (P< 0.001)。结论: ERCC2和 MGMT基因的表达之间可能具有内在的联系,共同参与细胞对 DNA损伤的修复,并形成耐药;而 mdr1与上述两者之间不具有这种关系。

Abstract:Objective: This study was designed to evaluate the relationship among mdr1, ERCC2,and MGMT expression in human glioma. Methods: RT- PCR method was used to determine the mRNA levels of mdr1, ERCC2, and MGMT gene in 14 normal brain tissues and 56 human gliomas,and the relationship between these gene expression was analyzed. Results: In normal brain tissues, there was significant concordance between ERCC2 expression and mdr1 (P< 0.01) expression or MGMT (P< 0.001) expression. A significant correlation was also found between the expression of ERCC2 and MGMT in glioma specimens (P< 0.001). Conclusion: The results suggest that there may be intrinsic relationship between ERCC2 and MGMT expression in human gliomas which affect DNA repair resulting in drug resistance.
