大承气颗粒药物血清对小鼠肠上皮内淋巴细胞产生/分泌IL-2的作用

摘    要:目的:探讨大承气颗粒药物血清对小鼠肠上皮内淋巴细胞产生/分泌白细胞介素-2(interleukin-2,IL-2)的作用.方法:以不同浓度的大承气颗粒剂灌胃大鼠后不同时间点于无菌条件下采取腹腔静脉血,制备药物血清;分离培养肠上皮内淋巴细胞(intestinal intraepithelial lymphocytes,IELs),以药物血清作用于IELs,设培养液对照、IEL对照、正常血清对照,采用放射免疫分析法测定IL-2.结果:不同浓度大承气颗粒的药物血清在多数时间点的作用明显强于培养液对照、IEL对照及正常大鼠血清对照.正常大鼠血清对照组与IEL对照组的IL-2水平很接近.同时间点的药物血清比较,在30 min和12 h时,4%大承气颗粒的药物血清作用强;在3 h时,40%大承气颗粒的作用最强.4%、20%大承气颗粒的药物血清使小鼠分泌IL-2的作用在1.5 h达峰值,在24 h出现第二个峰值;40%大承气颗粒药物血清使IEL分泌IL-2的峰值在3 h.结论:不同浓度的大承气颗粒制备的药物血清具有使IEL产生/分泌IL-2的作用;该作用涉及到药物本身和药物在体内的活性代谢物和/或其诱导的内源性活性物质的作用.

Effect of Drug Sera from per os Da Cheng Qi(大承气) Granule on Production/Secretion of IL-2 by Intestinal Intraepithelial Lymphocytes of Mice

Abstract:Objective To explore the effect of drug sera from mice per os Da Cheng Qi granules on production/secretion of interleukin(IL-2) by intestinal intraepithelial lymphocytes of mice Methods Drug sera were prepared through blood collected from the inferior vena cava of mice under sterility, condition at various time after per os Da Cheng Qi granules solution of different concentrations. Intestinal intraepithelial lymphocytes(IELs) in mice were separated and cultured,and then treated by the drug sera; control groups were set simultaneously. Including media control, IELs non-treated control, control of IELs treated by sera from normal rats; IL-2 was determined with radioimmuoassay. Results IL-2 production/secretion of IELs treated with drug sera from mice per os Da Cheng Qi granules of various concentrations and from blood collected at more points of time increased significantly than that of media control, IELs non-treated control, control of'IELs treated with sera from normal mice. IL-2 level of IELs treated with sera from normal mice was similar to that of IELs non-treated control. With drug sera from blood collected at the same time, IL-2 production/secretion of IELs treated with drug sera of mice per os 4% Da Cheng Qi granules was the most significant at 30 min and 12 h; so was that of 40% Da Cheng Qi granules at 3 h, IL-2 production/secretion of IELs treated with drug sera of mice per os 4% and 2% Da Cheng Qi granules peaked at 1.5 h, the second peak occurred at 24 h; so did that of 40% Da Cheng Qi granules at 3 h. Conclusion Drug sera from rats per os Da Cheng Qi granules of various concentrations are able to make mice IELs produce/secrete IL-2, this effect may relate to the action of Da Cheng Qi granules and components and their active metabolites and/or induced intrinsic substances induced by them.
