放射免疫法测定尿中甲基苯丙胺

目的:建立一种准确可靠、简易快速地定量甲基苯丙胺(MA)的检测方法。方法:采用碳化二亚胺法制备甲基苯丙胺半抗原,免疫兔使其产生相应的抗体与Na~(125)I标记抗原组成试剂盒;通过对健康志愿者和中枢兴奋剂滥用者的尿样检测,进行试剂盒临床应用的预试验。结果:试剂盒的灵敏度:1ng·ml~(-1),特异性:与MA有100％的反应,与相关的药物交叉反应小或无交叉反应。完成735份正常人和58份中枢兴奋剂滥用者的尿样分析;99％正常人尿中非特异干扰(相当于)甲基苯丙胺在500ng·ml~(-1)以下,单纯冰毒吸食者在末次吸毒后24h甲基苯丙胺含量>1000ng·ml~(-1),最大达7000ng·ml~(-1)以上。阈值定为500ng·ml~(-1)。结论:用放免法测定尿中甲基苯丙胺的~(125)I-MA试剂盒,灵敏度高,特异性好,结果精确,操作简便,是较常用的一种定量分析MA的有效方法。

DETERMINATION OF METHAMPHETAMINE IN URINE BY RADIOIMMUNOASSAY

Objective: To set up a quantitation method for detecting methamphetamine(MA) in urine. Methods: (1 )The N - 4 - aminobutylmethamphetamine - BSA conjugate was prepared by a carbodiimide method, and were immunized rabbits with it to produce an antibody to MA.125I - methaphetamine was used as a labeled compound for radioimmunoassay(RIA). (2)The preliminary trial for clinical application of 125I - MA kit was conducted. Results: Sensitivity:MA 1 ng ml-1, sepecificity: MA 100% . There was little or no cross -reaction on 18 related compounds. The samples in urine of 735 cases of nonnal population and 58 cases of central stimulant abuseres were detected by RIA . The ME of ( results expressed as ME : methamphetamine equivalents) more than 99% of normal samples was under 500 ng ml-1 .In contrast,the ME concentration in the urine of stimulant abusers 10 h - 34 h after the last administration exceeded 1000 ng ml-1 . Cut off value: 500 ng ml-1 . Conclusion: A inghly sensitive radioimmunoassay method for detecting MA in urine is set up. It can reliably quantitate MA in urine rapidly.
