结直肠癌组织乳腺癌易感基因1与表皮生长因子受体的蛋白表达及其临床意义

目的：探讨乳腺癌易感基因1（ BRCA1）和表皮生长因子受体（ EGFR）蛋白在结直肠癌组织中的表达及其临床意义。方法收集结直肠癌标本80例、腺瘤标本20例、正常黏膜标本20例，采用免疫组化法检测BRCA1和EGFR蛋白的表达情况，比较不同临床病理特征结直肠癌患者的癌组织BRCA1蛋白、EGFR蛋白的阳性表达率，分析结直肠癌患者的癌组织BRCA1蛋白、EGFR蛋白表达的相关性。结果（1） BRCA1蛋白的阳性表达率在结直肠癌组织低于腺瘤及正常黏膜组织（P＜0．05），有淋巴结转移患者其BRCA1蛋白阳性表达率也低于无淋巴转移患者（P＜0．05）。（2）EGFR蛋白阳性表达率在结直肠癌组织高于腺瘤及正常黏膜组织（P＜0．05）；肿瘤直径＞5 cm、中（低）分化腺癌、Duke C＋D期、有淋巴结转移的患者EGFR蛋白阳性表达率更高（P＜0．05）。（3）结直肠癌组织中BRCA1与EGFR蛋白表达呈负相关（r＝－0．219， P＜0．05）。结论 BRCA1及EGFR蛋白在结直肠癌的异常表达，对肿瘤的发生发展起到重要作用。

Protein expressions and clinical significance of breast cancer susceptibility gene 1 and epidermal growth factor receptor in colorectal cancer tissue

Objective To investigate protein expressions and clinical significance of breast cancer susceptibility gene 1(BRCA1) and epidermal growth factor receptor ( EGFR) in colorectal cancer tissue .Methods Eighty specimens of colorectal cancer ,20 specimens of colorectal adenoma ,and 20 specimens of normal colorectal mucosa were collected .The expressions of BRCA 1 and EGFR proteins were detected by immunohistochemistry method .The positive expression rates of BRCA 1 and EGFR proteins in cancer tissue were compared between the patients with different clinicopathological features .The correlation between BRCA 1 and EGFR protein expressions were analyzed in the patients with colorectal cancer .Results ①The positive expression rate of BRCA 1 protein in colorectal cancer tissue was lower than in adenoma tissue or normal mucosa tissue (P<0.05),and so in patients with lymphatic metastasis than in patients without lymphatic metastasis(P<0.05).②The positive expression rate of EGFR protein was higher in colorectal cancer than in adenoma tissue or normal mucosa tissue(P<0.05),and so in the patients with tumor diameter >5 cm,moderately(poorly)differentiated adenocarcinoma, Duke stage C+D,and lymphatic metastasis(P<0.05).③The expression of BRCA1 protein negatively correlated with the expression of EGFR protein in the colorectal cancer tissue (r=-0.219,P<0.05).Conclusion There exist abnormal expressions of both BRCA 1 and EGFR protein,which may play an important role in colorectal tumorigenesis and development .
