摘    要:目的:探讨4种外源性脂肪酸对乳腺癌细胞增殖的影响及其作用机理。方法:在体外实验中实用四甲基偶氮唑蓝法观察外源性脂肪酸对乳腺癌细胞增殖的影响,并对细胞脂质过氧化反应产物丙二醛含量进行测定。结果:多不饱和脂肪酸可抑制乳腺癌细胞增殖,饱和脂肪酸及单不饱和脂肪酸作用不明显。4种脂肪酸对成纤维细胞没有明显抑制作用。多不饱和脂肪酸可引起细胞内丙二醛含量升高。结论:多不饱和脂肪酸可选择性地抑制乳腺癌细胞增殖,脂质过氧化反应增强可能是其主要作用机理。

Abstract:Objective To observe the effect of fatty acids on the growth of breast cancer cells and to explore its mechanisms. Methods Fatty acids were added to cultured breast cancer cell line MCF_7.The growth of cancer cells were determined by 5_diphenyl tetrazolium bromide (MTT) assay. The intracellular content of malonaldehyde (MDA) was measured and used to indicator of lipid peroxidation. Results The growth of MCF_7 cells was inhibited by polyunsaturated fatty acids (PUFAs). But saturated fatty acids and monounsaturated fatty acids did not show inhibitory effect on MCF_7 cells. PUFAs caused significant rising of intracellular MDA content. Conclusion PUFAs could inhibit the growth of breast cancer cells. The induction of Lipid peroxidation may be mechanism.
