摘    要:目的:研究非病毒载体法基因转染的最适条件及其潜在价值。方法:利用脂质体介导和配体定向法将带有野生型P^53基因(Wt-P^53)cDNA的质粒引入细胞,用直接镜检、软琼脂培养、四甲基偶氮唑(MTT)法和乳酸脱氢酶法等研究细胞生长特性的改变情结果:绝大多数细胞被转染,肿瘤细胞生长受到明显抑制而发生凋亡。结论:本方法对于基因体外转染是可行的,并且有可能将来用于小细胞肺癌的P^53基因治疗。

Abstract:Objective:To optimize the P 53 gene transfection conditions and value the nonviral gene vector methodMethods:The plasmids with wildtype P 53 gene (WtP 53 ) were liposomemediated,ligandtargeted,and introduced into cancer cells,then the comparative studies were made with direct microscopy,soft agar culture,monotetrazolium (MTT) analysis and lactate dehydrogenase (LDH) testResults:Most cells were transfected,the growth of which was apparently arrested while apoptosis was observedConclusion:The gene transfection protocol was practical in vitro and might be useful in P 53 gene therapy for small cell lung cancer
