双氯灭痛缓释片的初步研制

摘    要:以壳聚糖作为缓释剂,通过湿颗粒压片法,制备了双氯灭痛缓释片。采用萃取紫外法,测定了家兔单剂量口服片(液)后血清中的双氯灭痛浓度,经计算机嵌合求得药动学参数,结果表明:缓释片的体内过程符合一室模型,并且体内外相关性良好,消除半衰期显著延长。

Abstract:Using claitosan as sustained release agent, the diclofenac sodium sustained release tablet (DSSR) by wet-granule compressing was prepared. The serum concntration of diclofenac was measured by extraction-ultraviolet spectroscopy after oral administration of single dose in rabbits, and the pharmacokinetics parameter was obtained by computer inset. The results indicated that the DSSR was according with unimol model in vivo. It was appropriate of the correlation between absorption in vivo and release in vitro. The elimination half-time was obviously prolonged.
