阿斯匹林对胃癌细胞株SGC-7901的细胞毒作用及体外增效作用

目的：观察阿斯匹林（ASA）对胃癌细胞株SGC－7901的细胞毒作用以及联用其它抗肿瘤药的增效作用。方法：应用流式细胞仪（FCM）测定细胞周期，噻唑蓝（MTT）法测定药物细胞的抑制率。结果：MTT显示体外ASA对SGC－7901有细胞毒作用，与ASA的浓度和作用时间有显著的相关性。同时可使SGC－7901细胞周期中S期及G2／M期比例升高，G1期比例下降，呈一定的剂量效应关系。低浓度ASA与5－氟脲噻啶（5－Fu)、阿霉素（DOX）和丝列霉素（MMC）合用，可增强它们对SGC－7901的杀伤作用，与各药物有相加或协同作用。结论：ASA体外对SGC－7901有细胞毒作用，可明显阻断SGC－7901细胞在S期和G2／M期；增强上述药物对SGC－7901的杀伤作用。鉴于所试药物对细胞周期的影响不同，提示ASA的体外增效作用可能与此相关。

The effect of aspirin on the SGC-7901 cellcycle and its enhance in vitro cytotoxicity by anticancer drugs

Objective To study the cytotoxic effects of aspirin on the SGC 7901 cell and the synergism of cytotoxic effects when combined with other anticancer agents.Methods Cell cycle was determined using Flow Cytometry (FCM) analysis,Cytotoxicity assay was tested by MTT method.Results MTT showed that the growth of SGC 7901 was inhibited by aspirin.The inhibitory rates for growth of SGC 7901 were positively correlated with the concentration and duration of aspirin in the media.The FCM data showed that the cell cycle pattern of SGC 7901 cells was changed when treated with aspirin.After exposure of the cells to various concentration of aspirin for 24h,the S and G 2/M phase ratio was elevated significantly in a dose dependent increase.The inhibitory rates of aspirin combined with 5 Fu or MMC or DOX were higher than that of these drugs used alone.Conclusion The results indicated that aspirin inhibited the growth of human gastric cancer cell line SGC 7901 or DOX were higher than that of these drugs used alone.The results indicated that aspirin inhibited the growth of human gastric cancer cell line SGC 7901 cells in S and G 2/M phase.The aspirin could enhance the cytotoxicity of the drugs mentioned above.The different effect of these agents on the various phases of cell cycle kinetics may be partially explained the enhanced effect of aspirin.
