雷米芬太尼的依赖性潜力

目的··:评价雷米芬太尼 (Rem)的依赖性潜力。方法·· :采用大、小鼠催促戒断模型、大鼠自然戒断模型、大鼠条件性位置偏爱模型和药物辨别模型进行评价。结果·· :(1)在催促戒断试验中 ,小鼠没有出现明显的戒断反应 (P>0.05);(2)大鼠催促戒断试验中 ,Rem在出现死亡的剂量下 ,只观察到轻微的戒断症状(P<0.05);(3)在自然戒断试验中 ,大鼠没有出现明显的戒断反应;(4)在条件性位置偏爱试验中 ,Rem5.0μg·kg-1形成了明显的条件性位置偏爱效应;(5)在大鼠药物辨别试验中 ,Rem20μg·kg-1 能完全替代大鼠对2.5mg·kg-1 吗啡的辨别效应。结论·· :Rem具有产生精神依赖性和身体依赖性的潜力 ,但由于其作用时间短 ,较难形成身体依赖性 ,而其精神依赖性的形成 ,不受药物作用时间的影响

DEPENDENCE- PRODUCING POTENTIAL OF REMIFENTANIL

Objective: To evaluate the dependence_producing potential of remifentanil. Methods: Mice jumping test, naloxone-precipitated and spontaneous withdrawal models in rats, conditioned place preference and drug discrimination in rats were adopted. Results: (1) In mice jumping test, the mice didn't show any withdrawal symptoms;(2)The remifentanil-treated rats only manifested very light withdrawal symptoms precipitated by naloxone;(3) In the spontaneous withdrawal experiment, the remifentanil-treated rats didn't show significant loss of body weight after remifentanil withdrawal;(4)In the conditioned place preference test, rats developed place preference effect at the dose of 5.0 μg·kg-1 remifentanil;(5) In the drug discrimination experiment, 20 μg·kg-1 remifentanil completely substituted for the discriminative effect of 2.5 mg·kg-1 morphine. Conclusion: Remfentanil has drug dependence-producing potential. However, it is very difficult for remifentanil to develop physical dependence due to its ultra-short action , but the development of psychic dependence may be independent of duration of drug action.
