摘    要:目的研究鼠龄对骨髓间充质干细胞(MMSCs)增殖特性的影响.方法用贴壁培养法分离纯化不同鼠龄的MMSCs,传代扩增,测定生长曲线,形态学观察,免疫组化鉴定,电镜观察MMSCs的超微结构.结果2月龄大鼠MMSCs集落形成率、增殖能力显著高于6月龄大鼠,细胞化学染色CD44、CD105均阳性,电镜支持幼稚细胞.结论 2月龄大鼠比6月龄大鼠更适于MMSCs的提取.

Abstract:Objective To investigate the effect of age on the proliferation of rat marrow mesenchymal stem cells(MMSCs).Methods MMSCs were isolated and purified with adherence plasticity method.The morphologic features and growth status by light and electron microscopies were observed.The cell surface antigens by immunochemical technique was examined.Results The ratio of clone forming unit and proliferation capacity in rat of two months were significantly higher than those of six months (P<0.05).CD_ 44 and CD_ 105 were positive in MMSCs by immunohistochemistry technique.These cells were naive cells by electron microscopies.Conclusion Rats of two months were more appropriate than those of six months for MMSCs extraction.
