摘    要:他汀类药物作为降低胆固醇的主要药物,具有稳定动脉斑块、抗炎和抗氧化作用,临床应用广泛。近年来,许多基础及临床研究表明,他汀类药物可通过抑制甲羟戊酸途径,促进骨形态发生蛋白-2基因表达;阻止脂肪细胞生成,同时使骨髓基质干细胞向成骨细胞分化;调节核因子(NF)-κB通路,抑制NF-κB受体激活剂所诱导的破骨细胞形成等多种作用机制影响骨代谢,从而使他汀类药物成为骨质疏松症药物治疗的研究热点。

Abstract:As the major kind of medicines in decreasing cholesterol, statins have been widely used to decrease LDL-cholesterol. Recently, many basic and clinical studies had indicated that statins might influence bone metabolism by many mechanisms including inhibiting mevalonate pathway, enhancing gene expression of bone morphogenetic protein-2, inhibiting the formation of adipocyte, simultaneously stimulating bone marrow-derived mesenchymal stem cells to differentiate into osteoblasts, and inhibiting receptor activator of nuclear factor-kappaB(NF-κB) ligand induced the formation of osteoclasts by mediating NF-κB pathway, therefore statins become the focus drugs in the experimental study of osteoporotic medication.
