Ras基因在肝癌中表达的意义

目的：探讨ras基因家族在肝癌中表达的意义。方法：应用原位杂交与免疫组织化学方法检测肝癌组织与癌旁肝病组织中ras基因蛋白p21及ras家族基因N-ras,H-ras和Ki-ras。结果：肝癌与癌旁肝病组织中ras基因阳性率分别为90％（36／40）和82．3％（28／34），相对正常组织中ras基因阳性率为33．3％（2／6），癌与癌旁肝病组织中ras基因阳性率无显著差异，而肝癌、癌旁肝病组织与相对正常肝组织中ras基因阳性率有显著差异（P＜0．01）。且原位杂交方法与免疫组化方法检测ras基因表达无显著差异，原位杂交检测的ras基因家族成员之间亦无显著差异，但ras表达强度与癌分化程度有关，呈抛物型特点。结论:ras基因的表达是肝癌发生的早期事件，是肝癌形态学发生中的分子改变，癌变过程癌细胞合成增加，但在癌分化降低时，活跃增殖的癌细胞中ras合成反而受抑，免疫组化法较原位杂交方法检测ras基因更经济、简便。

SIGNIFICANCE OF Ras GENE EXPRESSIONS IN HEPATOCELLULAR CARCINOMA

Objective:To study the significance of the ras gene expressions in Hepatocellular Carcinoma.(HCC).Method:The expressions of N-ras p21 protein and the oncogenes of ras family(N-ras,H-ras,Ki-ras) in HCC and paracarcinoma tissues,including active hepatitic cirrhosis and normal liver tissues were studied by immunohistochemistry and in-situ hybridization.Results: the positive rates of ras gene in HCC and paracarcinoma disease were 90%(36/40) and 82.3%(28/34),respectively.The positive expressions were not statistically between immunohistochemistry and in-stiu hybridization and in ras family genes different in those groups but were related to the degree of malignant differentiation (P<0.01).Conclusion: Our data indicate that the expressions of ras gene is an early event in HCC,also being one of the molecular changes in morphogenesis of HCC.Ras gene detected by immunohistochemistry were superior to that in-situ hybridization.
