小檗碱对颈动脉粥样硬化家兔颈动脉病理改变和VCAM-1转录水平的影响

摘    要:目的:探讨小檗碱预防颈动脉粥样硬化形成的作用机制。方法:将24只雄性新西兰大白兔随机分为对照组、模型组和小檗碱组各8只。对照组给予普通饮食;模型组和小檗碱组给予高脂饲料喂养1周后行右侧颈动脉内膜空气干燥术,术后,前者继续高脂饲料喂养4周以制成颈动脉粥样硬化模型,后者在前者的基础上每日灌服小檗碱(100 mg/kg)。第5周麻醉处死,取右侧颈动脉组织,行HE染色观察颈动脉病理改变,采用RT-PCR法测定VCAM-1 mRNA含量。结果:与对照组比较,模型组颈动脉内膜明显增生,内膜下可见大量泡沫细胞堆积,有明显粥样硬化斑块形成;而小檗碱组内膜轻度增厚,内膜下仅有少量泡沫细胞。模型组、小檗碱组和对照组VCAM-1/-βactin扩增带吸光度比值依次降低,差异有统计学意义(0.61±0.11,0.20±0.04,0.81±0.11,P<0.01)。结论:小檗碱可能通过抑制VCAM-1的表达预防颈动脉粥样硬化的形成。
