持续母源性经口暴露PBDE-209对其子代大鼠体液免疫毒性影响

摘    要:  目的  观察三七免疫毒性,为评价其安全性提供依据。  方法  采用产自云南文山州的三七作为受试物,机械粉碎成粉末,过120目网筛后供试验使用。40只SD雄性大鼠随机分为对照组和低、中、高剂量三七组(0.750、2.372、7.500 g/kg),每组10只,连续灌胃给予受试物90 d,定期观察一般健康状况,每周记录体重及摄食量;检测血液学、生化指标、观察组织病理学改变,采用血清溶血素试验、NK细胞活性试验、淋巴细胞转化试验及外周血淋巴细胞活化水平试验,评价受试物对SD大鼠免疫功能的影响。  结果  各组大鼠一般生理体征、行为、外观、皮毛和大小便等均无异常;与对照组比较,各剂量三七组大鼠体重、白细胞计数、白细胞分类计数,血清总蛋白、白蛋白、谷丙转氨酶及谷草转氨酶,脾、胸腺的重量与系数,组织病理学检查均无明显改变;与对照组比较,各剂量三七组大鼠半数溶血值(HC50)、NK细胞活性、淋巴细胞转化能力均无明显变化;与对照组比较,低、中、高剂量三七组大鼠NK细胞表面标记(CD161α+CD25+)[分别为(10.17 ± 5.20)、(7.25 ± 3.22)、(8.63 ± 1.90)]、B细胞表面标记(CD45RA+CD25+)[分别为(9.64 ± 4.83)、(7.76 ± 3.18)、(9.06 ± 2.85)]、T细胞表面标记(CD3+CD25+)[分别为(17.08 ± 4.62)、(14.11 ± 2.87)、(14.27 ± 3.31)]均无明显变化。  结论  在本实验条件下,三七对SD大鼠免疫功能无明显影响,无免疫毒性。

Influence of dioxide titanium nanoparticles on immune system indicators in rats

Abstract:  Objective  To observe the immune toxicity of Panax notoginseng (Pn) and to provide a basis for safety assessment of Pn.  Methods  We collected Pn samples in Wenshan of Yunnan province and triturated them into powder and screened with a 120 mesh sieve before experiment. Fourty male Sprague-Dawley (SD) rats were randomly divided into four groups (10 in each group): a control group and low, moderate, and high Pn groups treated with gastric gavage of Pn at doses of 0.750, 2.372, and 7.500 g/kg body weight once a day continuously for 90 days. All the rats were observed for general conditions and their body weight and feed intakes were recorded periodically and hematologic and biochemical indicators were detected and pathological changes in spleen and thymus tissues were observed at the end of experiment. The immunologic function of the rats was assessed with tests of serum hemolysin, natural killer (NK) cell activity, lymphocyte transformation, and peripheral blood lymphocyte activation.  Results  No abnormalities in physical signs and behaviors were observed among all the rats. Compared to those of the control rats, no significant differences were detected for the rats of three Pn groups in body weight, white blood cell count, white blood cell differential count, total serum protein, serum albumin, alanine aminotransferase, aspartate aminotransferase, 50% hemolysin value, NK cell activity, lymphocyte transformation, and weight and organ coefficients of spleen and thymus. There were also no obvious pathological changes in spleen and thymus tissues in the rats of Pn groups. For rats of low, moderate and high Pn groups, the expression of CD161α+CD25+ in NK cells (10.17 ± 5.20, 7.25 ± 3.22 and 8.63 ± 1.90), CD45RA+CD25+ in B lymphocytes (9.64 ± 4.83, 7.76 ± 3.18 and 9.06 ± 2.85), and CD3+CD25+ in T lymphocytes (17.08 ± 4.62, 14.11 ± 2.87 and 14.27 ± 3.31) were not significantly different from those for the rats of control group.  Conclusion  The experiment results demonstrate no immunotoxicity of Panax notoginseng in SD rats.
