刺五加注射液对大鼠肝缺血再灌注损伤的保护机制

摘    要:目的探讨刺五加注射液对大鼠肝缺血再灌注损伤的保护机制作用。方法将雄性Wistar大鼠30只随机等分为假手术组(对照组)、缺血再灌注组(模型组)、刺五加注射液组(100mg/kg)。检测血清中转氨酶(AST、ALT)、超氧化物歧化酶(SOD)、髓过氧化物酶(MPO)、丙二醛(MDA)水平、肝组织中超氧化物歧化酶(SOD)、丙二醛(MDA)、钙一三磷酸腺苷酶(Ca2+ -ATPase)含量,利用免疫组化法检测B细胞淋巴瘤/白血病-2(B—celllym-phoma/leukemia-2,Bcl-2)、Bax蛋白的表达。结果与模型组比较,刺五加注射液组有效降低血清中ALT、AST及MPO的含量(P〈0.01);明显提高血清及肝组织中SOD的活性(P〈0.01),降低MDA的含量(P〈0.01);明显提高肝组织中ca2+ -ATPase的活力(P〈0.01);增加肝组织中Bcl-2蛋白的表达(P〈0.01),减少肝组织中Bax蛋白的表达(P〈0.01);上述差异均有统计学意义。结论刺五加注射液可通过增强自由基清除能力,抑制中性粒细胞聚集、细胞内钙超载及细胞凋亡来有效减轻肝损伤,对肝缺血再灌注损伤具有明显的保护作用。

Protective effects of Acanthopanax senticosus injection on hepatic ischemia- reperfusion injury in rats

Abstract:Objective To discuss the protective effects of Acanthopanax senticosus injection on hepatic ischemia- reperfu- sion injury in rats. Methods Thirty adult male Wistar rats were randomly divided into 3 groups : Sham - operated group, control (HIRI) group,Acanthopanax senticosus injection( 100 mg/kg) group. Serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) level, superoxide dismutase (SOD) activity and myeloperoxidase (MPO) and maleic dialdehyde (MDA) content were measured. Superoxide dismutase (SOD) activity, content of MDA and Ca2+ - adenosine triphosphatase (Ca2+ -ATPase) in liver tissue were measured. Expression of Bcl- 2 and Bax protein in hepatic tissue were detected with the immunohistochemistry methods. Results Compared with control group:serum ALT and AST and MPO content ( P 〈 0.01 ) and content of MDA ( P 〈 0.01 ) were significantly reduced and Activity of serum SOD ( P 〈 0. 01 ) was increased in group of Shuxuening; Also SOD(P 〈 0.01 ) activity in liver tissue was significantly increased and content of MDA(P 〈0. 01 ) and Ca2+ -ATPase(P 〈 0. 01 ) were significantly reduced in group of Acanthopanax senticosus injection; The expressions of Bcl - 2 protein were significantly increased and the expressions of Bax protein were significantly reduced in group of Acanthopanax senticosus injection in hepatic tissue ( P 〈 0. 01 ). Conclusion Acanthopanax senticosus injection can effectively reduce the hepatic injury, strengthen to the free radical scavenging capacity and inhibit neutrophils gathered,intracellular calcium overload and cell apoptosis. It has significant protective effects of hepatic ischemia - reperfu- sion injury in rats.
