热休克蛋白70对大鼠心肺复苏后神经细胞凋亡的影响

摘    要:目的:研究热休克蛋白70(HSP 70)对大鼠心肺复苏后神经细胞凋亡的影响,探索HSP对脑复苏的可行性。方法:将46只成年Wistar大鼠随机分为假手术组、模型组与干预组,假手术组不进行模型制作及治疗干预;模型组仅制作心搏骤停(SCA)并心肺复苏动物模型,不进行治疗干预;干预组大鼠在SCA并心肺复苏模型制作后静脉注射HSP 70。分别于6、12 h后检测各组大鼠脑海马区神经细胞凋亡数目、Bcl-2蛋白表达量,对比分析各组大鼠神经细胞凋亡情况的差异,验证HSP 70对大鼠心肺复苏后神经细胞凋亡的保护作用。结果:HSP 70对大鼠心肺复苏后神经细胞具有保护作用,能减少神经元细胞凋亡数目,增加Bcl-2蛋白表达。结论:HSP 70对大鼠心肺复苏后神经细胞有保护作用,对脑复苏具一定的治疗前景。

Abstract:Objective: To investigate the effect of heat shock protein(HSP) 70 on neuronal apoptosis after cardio-pulmonary resuscitation in rats.To explore the feasible prospect of heat shock response on cerebral resuscitation.Methods: Forty-six Wistar rats were randomly divided into the sham operation group,the model group and the treatment group.The sham operation group received neither SCA model made nor further treatment,the model group received SCA model made but not HSP 70 injection and the treatment group received both.The count of neuronal apoptosis and expression of Bcl-2 protein were measured respectively at 6,12 h after injection HSP 70.To contrast the differences of neuronal apoptosis among groups in order to verify the protective effect of HSP 70 on neuron after cardio-pulmonary resuscitation in rats.Results: HSP 70 had the effects of reducing counts of neuronal apoptosis and increasing expression of Bcl-2 protein after cardio-pulmonary resuscitation in rats.Conclusion: HSP 70 has the effect of preventing neronal cell from apoptosis after cardio-pulmonary resuscitation in rats and has the feasible prospect on cerebral resuscitation.
