摘    要:目的观察黄芪醇提有效部位在病毒性心肌炎体外细胞培养模型中的药效作用。方法柯萨奇病毒感染乳鼠原代心肌细胞后不同时间加入黄芪总提取物和/或病毒,逐日观察心肌细胞搏动次数,细胞病变、MTF染色比较各组间活细胞数的变化,病毒繁殖抑制实验比较TCID50的差别以验证黄芪总提取物的抗病毒活性。实验以盐酸胍作为阳性对照药。结果黄芪醇提有效部位在体外实验中能有效抑制CVB3引起的细胞病变(P〈0.01),加药组比病毒对照组TCID50降低两个以上对数值。结论黄芪总提取物对体外培养的心肌细胞具有保护作用。

Abstract:Objective To investigate the antiviral effect of Total Extract of Astragalus to cultured cardiac myocytes infected with cosackievirus B3. Methods Cardiac myocytes were derived from newborn Sprague-Dawlay rat heart. The cells were cultured for 3 days and then treated with CVB3 and/or TEA. Cytopathic effect and beat of cardiac myocytes was observed with inverted phase contrast microscope and cell viability was evaluated with MTT assay. TCID50 was used to evaluate the rcinfectious effects of the used CVB3 in the cultured media on cultured cardiac myocytes. Guanidine Hydrochloride was used as positive control. Results TEA can effectively lessen the occurrence of cytopathic effect (P 〈 0.01 ). 10 μg/ml of TEA significantly suppressed infectious virus propagation and reduced the TCID50 from 6.8 to 4.3. The antiviral effect of TEA was better than Guanidine Hydrochloride. Conclusion TEA had a protective effect to cultured cardiac myocytes infected by CVB3.
