多重引物与共有引物PCR法检测宫颈癌组织中的HPVDNA的比较及意义

人子宫颈鳞状上皮癌(简称宫颈癌)是妇女死亡的主要原因之一,近年来,实验室的研究证明了某些类型的人乳头瘤病毒(Human Papillomavirus,HPV)与宫颈癌以及其它几种生殖肿瘤病有明显的关系。本文应用多重引物与共有引物PCR法对山西60例宫颈癌活检组织人乳头瘤病毒的检测,结果表明,多重引物PCR检出率为81.66％;共有引物PCRHPV检出率为85％,两法符合毕为93.33％。共有引物PCR法比多重引物PCR法检出率高。两种方法各有其特点。本文认为大规模普查最好采用共有引物PCR法,它的特点是广谱,费用低。对临床HPVs感染的鉴别诊断最好选用多重PCR法。

Multiple Primer and Consensus Primer PCR Methodsfor Dection of Human Papillomavirus in Cervical CarcinomaTissue of Woman and Clinical Significance

The squamous cell carcinoma of uterine cervix (cervical cancer) is one of the leading causes of female death from cancer. Laboratory studies over the past decade have established a strong association between high risk human papillomavirus (HPV) and cervical cancer and several other anogenital carcinomas. Consensus primer and multiple primer PCR methods have been successfully established by our laboratory firstly in China. Using consensus primer and multiple primer PCR meth ods, we detected human papillomavirus DNA of 60 cases cervical carcinoma biopsy tissues and comparison of two methods for detection of HPV were carried out. The coincident rate of consensus primer and multiple primer PCR was 93. 33%. The differences of two methods were not statistically significant (p>0. 001). But two methods have respective characteristic. We consider that consensus primer PCR is an important assay for mass screening of high risk HPVs infected woman and multiple primer PCR may select to distinguish HPVs types from clinical infection of woman. Therefore through the comparison of multiple primer and consensus primer methods, two methods were proved tube stable, the result obtained is reliable.
