天麻素对氯化钾诱导的培养神经细胞释放谷氨酸的影响

摘    要:目的 考察天麻素对由氯化钾诱导的神经细胞释放谷氨酸的影响。方法 通过高效液相 (HPLC)测定人神经母细胞瘤SH SY5Y细胞系培养液中谷氨酸的含量。结果 当细胞外液中有钙离子时 ,随着氯化钾浓度的增加 (10~ 30mmol/L) ,释放到培养液中的谷氨酸的含量也相应增加 ;当培养液中去除钙离子时 ,细胞外液中谷氨酸的浓度不随氯化钾浓度的变化而改变。在含钙培养液中加入 4mg/ml的天麻素 ,再加入 10~ 5 0mmol/L氯化钾 ,未见细胞外液中的谷氨酸含量增高。结论 氯化钾诱导的SH SY5Y细胞谷氨酸的释放增多与细胞外液钙离子内流有关 ;天麻素可以颉颃由氯化钾诱导的神经细胞谷氨酸释放增加的作用。

Abstract:Objective To investigate the effect of gastrodin on the release of glutamate induced by KCl in cultured human neuroblastoma SH SY5Y cell line. Methods\ The concentration of glutamate in culture medium was measured by HPLC. Results\ (1)The concentration of glutamate in culture medium increased with the increment of the concentration of KCl (10 30mmol/L). The above change was not observed when there were no calcium ions in culture medium. (2) In the presence of gastrodin (4?mg/ml) in the medium, there was no increase in the concentration of glutamate in culture medium with the increment of concentration of KCl (10 50mmol/L). Conclusion\ KCl can induce the release of glutamate from cultured SH SY5Y cells, and this kind of release of glutamate can be antagonizecd by gastrodin.
