脊髓神经干细胞在Sapeptide材料上增殖与分化的研究

摘    要:目的观察脊髓神经干细胞在Sapeptide材料上的增殖和分化情况,并对分化的细胞进行一定功能检测,探讨其细胞假体在脊髓损伤中应用的可能性。方法观察塑形的Sapeptide材料,将培养的脊髓神经干细胞接种其上,然后鉴定干细胞并通过体积分数5%胎牛血清诱导分化,鉴定分化细胞的类型及比例,并与无材料的干细胞的分化情况相比较,最后对材料上的分化细胞进行缝隙连接蛋白32(CX32)和生长相关蛋白43(GAP-43)染色评价。结果Sapeptide材料在等渗盐水中可塑形,扫描电镜示具有规则的网状排列,脊髓神经干细胞在其上可增殖和分化,分化的细胞较无材料的差异无统计学意义(P〉0.05),并且具有大量的CX32和GAP-43阳性细胞。结论脊髓神经干细胞在Sapeptide材料上增殖和分化良好,假体在脊髓损伤的修复中可能具有潜在的应用前景。

Abstract:Objective To observe the proliferation and differentiation of neural stem cells from spinal cord on Sapeptide (Self-assembling peptide), detect the function of the differentiated cells so as to investigate the potential use of the mixture body in spinal cord injury (SCI). Methods Scanning electron microscope (SEM) was used to observe Sapeptide, to which neural stem cells identified by Nestin and BrdU were attached. Then, the neural stem cells were differentiated by 5% fetal bovine serum. The species and percent of the differentiated cells were examined to compare with those without Sapeptide. Finally, the differentiated cells were evaluated via immunohistochemistry stain by using connexin32 (CX32) and growth associated protein-43 (GAP-43).Results Sapeptide was shaped in 9 g/L NaCl showed regular net under SEM. Neural stem cells from spinal cord could proliferate and differentiate on the Sapeptide scaffold as well as had plenty of cells with CX32 and GAP-43, with no obvious difference compared with those free from Sapeptide (P>0.05). Conclusion Neural stem cells from spinal cord are well proliferated and differentiated on Sapeptide. The mixture body may have potential application in repair of SCI.
