复方藤梨根制剂对K562/ADM多药耐药作用的体内实验研究

摘    要:目的:研究复方藤梨根制剂逆转人红白血病耐药细胞株K562/ADM裸鼠移植瘤的多药耐药效应及机制。方法:建立K562/ADM裸鼠移植瘤多药耐药模型;以流式细胞仪(FCM)检测各组肿瘤细胞膜上P-gp的表达和肿瘤细胞内ADM蓄积浓度。结果:(1)ADM组P-gp的表达与NS组相比明显升高(P(0.05);不同剂量的FFTLG制剂与ADM合用时,P-gp的表达与ADM组相比显著下降(P(0.01)。单独应用FFTLG制剂,当浓度为0.8mg/mL时P-gp的表达与ADM组相比下降(P(0.05)。(2)与ADM组相比,ADM和不同剂量的FFTLG合用组肿瘤细胞内的ADM浓度均有上升(P(0.05)。结论:FFTLG制剂可以部分逆转荷瘤裸鼠的多药耐药性,其逆转效果似呈剂量依赖性。FFTLG制剂逆转荷瘤裸鼠多药耐药性的机理之一是通过下调肿瘤细胞膜上P-gp的表达或抑制P-gp的活性,增加肿瘤细胞内ADM的蓄积浓度而实现的。

Abstract:Objective:To study the effect and mechanism of Fufang TengliGen's reversal in multidrug resistant in nude mice of transplantation tumor of K562/ADM Cell.Methods:Nude mice of transplantation tumor of K562/ADM were established. Flow cytometry (FCM) was used to investigate the changes of P-gp expression and accumulation of intracellular adria mycine after different drug treatment.Results:(1) ADM group of P-gp expression compared with the NS group was significantly increased (P0.05). When different doses of FFTLG were combined with ADM,the P-gp expression compared with the ADM group was significantly decreased(P0.01). In FFTLG group,when the concentration was 0.8mg/mL,the P-gp expression compared with the ADM group was significantly decreased(P0.01). (2) Compared with the ADM group,the concentrations of ADM were increased in tumor cells group when different doses of FFTLG were combined with ADM(P0.05).Conclusion:FFTLG can effectively reversed the resistance of K562/ADM cells in nude mice and it seemed that the effect of reversal K562/ADM/nude was dependent on the dosage of FFTLG mixture.By down-regulation of tumor cell membrane expression of P-gp tumor cells to increase the volume concentration of ADM is the mechanism of FFTLG mixture to reverse multidrug resistance.
