复方藤梨根制剂下调乙酰肝素酶表达抑制胃癌细胞转移

目的：研究探讨复方藤梨根制剂抑制胃癌细胞转移的机制。方法：以SGC-7901胃癌细胞株建立胃癌裸鼠原位移植瘤模型,将裸鼠随机分为生理盐水组,复方藤梨根制剂低剂量、中剂量及高剂量组,接受连续21天口服灌注治疗。根据胃癌细胞转移的部位进行赋值评分,观察复方藤梨根制剂对裸鼠胃癌移植瘤转移的影响,并用荧光定量PCR仪（FQ-PCR）检测胃癌细胞乙酰肝素酶mRNA表达的影响。结果：根据胃癌细胞转移部位的赋值评分,生理盐水组与低剂量治疗组评分显著高于中、高剂量组;经FQ-PCR法检测,生理盐水组与低剂量治疗组的胃癌组织乙酰肝素酶mRNA表达显著高于中、高剂量治疗组。结论：在裸鼠动物模型,复方藤梨根制剂可能是通过下调乙酰肝素酶mRNA表达来抑制胃癌细胞的远处转移,发挥其抗肿瘤作用。

The Study of the Inhibit Effects of Metastasis By Fufang Tengligen Through Down-regulate Heparanase Expression in Gastric Cancer

Objective：To study and explore the inhibit effects and mechanisms of Fufang Tengligen mixture （FFTLM） on the metastasis of gastric cancer.Methods：The nude mice with the original gastric cancer transplant mode of SGC-7901 gastric cancer cell line were randomized into four groups,saline control group,low dose group of FFTLM ,middle dose group of FFTLM and high dose group of FFTLM,accepted 21 days of oral infusion treatment. The effects on the metastasis of gastric cancer were observed according to the score of the metastatic organ,and the expression of heparanase mRNA expression was detected by FQ-PCR technique.Results：Based on the score of the metastatic organ,the score of the saline control group and low dose group of FFTLM were markedly higher than that of middle and high dose group of FFTLM. After the detection by FQ-PCR technique,the heparanase mRNA gene expressions in saline control group and low dose group of FFTLM were greatly higher than that in middle and high dose group of FFTLM. Conclusion：Fufang Tengligen mixture can inhibit the metastasis of gastric cancer which is associate with the down-regulate the heparanase mRNA expression in nude mice model.
