MMP-2和TIMP-2神经端侧吻合修复神经损伤实验研究

摘    要:目的观察MMP-2和TIMP-2在神经端侧吻合部位表达的变化,探讨其在神经再生中的作用. 方法取36只新西兰兔,将右侧腓总神经切断并将其远侧端吻合至切断部位下方的正常胫神经,左侧做自身假手术对照.术后3,5,7,14,21,28 d灌注处死,用免疫组织化学方法检测端侧吻合部位MMP-2及TIMP-2的表达. 结果试验组MMP-2在损伤后5 d即出现表达升高,14 d达高峰,之后逐渐下降,28 d降至正常水平.TIMP-2的表达则于第3 d出现升高,21 d后出现下降,28 d降至正常;TIMP-2上升幅度与MMP-2相比较小.而自身假手术对照侧胫神经MMP-2、TIMP-2表达无明显变化. 结论神经损伤行端侧吻合术后,MMP-2和TIMP-2对轴突再生可能起重要作用.

Abstract:Objective To investigate the expression of MMP-2 and TIMP-2 at anastomotic site after sciatic nerver end-to-side neurorrhapy and to expound its role during nerve regeneration after peripheral nerve end-to-side neurorrhaphy. Methods It was used by nerve end-to-side neurorrhaphy model in New Zealand rabbit (cut common peroneal nerve from beginning and suture it to tibial nerve below cut site). Animals were randomly divided into two groups: normal control group and experimental groups. According to animal survival time,the latter were subdivided into 6 groups: 3,5,7,14,21,28 days postoperation. It has been detected the expression of MMP-2 and TIMP-2 at anastomotic site by immunohistochemistry method. Results It was found that expression of MMP-2 increase at 5 days group postoperation,and reach to the peak at 14 days group,then decrease gradually and reach to the normal level at 28 days group. But compared to control nerves,there is no significant change. The increase expression of TIMP-2 is similar to MMP-2 at 3 days group after end-to-side neurorrhaphy,then decrease to the normal level at 21 to 28 days group. There is no significant change at control group. It was lower that express of TIMP-2 compares to MMP-2. Conclusions These results indicate that MMP-2 and TIMP-2 play very important roles during nerve regeneration after peripheral nerve end-to-side neurorrhaphy.
