官腔脱落细胞中p53、ki-67、bcl-2、bax基因的表达及意义

摘    要:目的 采用实时荧光定量PCR法检测p53、ki-67、bcl-2、bax基因在子宫内膜癌及正常人官腔脱落细胞中的表达变化水平,探讨官腔脱落细胞中p53、ki-67、bcl-2、bax基因的表达变化在子宫内膜癌早期诊断中的意义.方法 收集健康志愿者及子宫内膜癌患者官腔脱落细胞样本共90例;提取细胞总RNA,采用实时荧光定量PCR法检测目的基因的表达变化水平;所有实验数据采用SPSS 17.0软件进行统计学分析.结果 荧光定量PCR结果显示,p53基因在正常人官腔脱落细胞中未检测到表达,随着恶性程度升高,表达量显著升高(P<0.01),ki-67的表达在正常人中表达量最低,随着恶性程度升高,表达量逐渐升高(P<0.05),bcl-2基因的表达逐渐降低(P<0.05),bax/bcl-2值随着恶性程度增高而降低.结论 p53、ki-67、bcl-2、bax基因的表达量在子宫内膜癌发生发展过程中呈一定的变化趋势,提示p53、ki-67、bcl-2、bax基因可作为子宫内膜癌早期诊断的分子标志物.

Abstract:Objective To dectect the expression of p53,ki-67,bcl-2and bax in intrauterine exfoliated cells of endometrial cancer and healthy women and to explore their clinical significance in the early diagnosing of endometrial cancer.Methods 80samples of intrauterine exfoliated cells of endometrial cancer patients and healthy people were collected and the total RNA were extracted,the target genes were detected by real-time quantitative PCR.Then compare the expression of p53,ki-67,bcl-2and bax in endometrial cancer patient and healthy people.All the experimental data were statistically analyzed by SPSS 17.0software.Results The P53gene was hardly checked out in normal people,but its expression level gradually increased with the degree of malignancy increased(P0.01).With the increasing degree of malignancy,the expression of ki-67was significantly increased(P0.05)but not bcl-2gene.The expression of bax gene was presented a negative correlation with bcl-2.Conclusions Real-time fluorescent quantitative PCR can sensitively and specifically detected the expression of P53,ki-67,bcl-2and bax in the intrauterine exfoliate cells.The P53,ki-67,bcl-2and bax can be used as molecular markers of endometrial cancer.
