β1整合素过表达促进体外角膜上皮细胞黏附和迁移

目的:探讨β1整合素过表达对角膜上皮细胞黏附和迁移的影响机制。方法:将β1整合素-GFP融合蛋白真核细胞重组表达质粒转染兔角膜上皮细胞,观察转染细胞的融合基因表达以及细胞的黏附和迁移能力。检测β1整合素转染对角膜上皮细胞粘着斑激酶(FAK)磷酸化的影响。结果:成功将β1整合素-GFP融合蛋白转染至兔角膜上皮细胞并使其过表达;β1整合素过表达能够明显增加角膜上皮细胞的黏附和迁移能力(P<0.05)并促进FAK磷酸化(P<0.05)。结论:β1整合素过表达能够明显促进角膜上皮细胞的黏附和迁移。

β1-integrin overexpression induces the ability of adhesion and migration of rabbit corneal epithelial cells

AIM:To investigate the effect of β1-integrin over-expression on the ability of adhesion and migration of rabbit corneal epithelial(RCE)cells.· METHODS:Eukaryotic expression vector encoding β1-integrin-GFP fusion DNA was transfected into RCE cells,the β1-integrin-GFP fusion gene was examined by RT-PCR and Western blotting.The adhesion to Matrigel and the migration of transfected cells were determined by adhesion assay and mobility assay.The phosphorylation of focal adhesion kinase(FAK)was examined by Western blotting.· RESULTS:RCE cells overexpressing β1-integrin-GFP fusion gene were successfully established.β1-integrin transfection significantly promoted the adhesive ability of RCE cells to Matrigel(P<0.05).β1-integrin overexpression promoted the migration ability and induced FAK phosphorylation in RCE cells(P<0.05).· CONCLUSION:These data suggest that overexpression of β1-integrin promote the ability of adhesion and migration of RCE cells,and FAK pathway may play an important role in this process.
