奥曲肽抑制人肝癌BEL-7402细胞生长的实验研究

摘    要:目的 探讨生长抑素类似物奥曲肽 (OCT)在体内对人肝癌BEL - 74 0 2细胞生长的影响。方法 建立人肝癌细胞株BEL - 74 0 2裸鼠移植瘤模型 ,实验组皮下注射奥曲肽 10 0 μg·kg- 1 ·d- 1 ,对照组给予生理盐水 2 0μl d ,给药 2 8天后处死裸鼠。取瘤 ,测量瘤重、瘤体积后 ,检测移植瘤组织中血管内皮细胞生长因子 (VEGF)、微血管密度 (MVD)的表达。结果 奥曲肽组瘤重、瘤体积与生理盐水组间有显著性差异 (P <0 .0 5 )。奥曲肽组VEGF的表达明显低于生理盐水组 (P <0 .0 5 ) ,而MVD在奥曲肽组的表达虽然低于生理盐水组 ,但统计学差异无显著性 (P =0 .0 5 5 )。结论 生长抑素类似物奥曲肽能够抑制裸鼠人肝癌移植瘤的生长 ,并能抑制瘤组织中VEGF的表达 ,同期移植瘤组织中微血管密度呈下降趋势。

Abstract:Objective To investigate the depressive effect of octreotide on the growth of human hepatocellular carcinoma BEL-7402 cell line in vivo. Methods The animal model of xenotransplanted hepatocellular carcinoma was established with BEL-7402 cell line in 12 nude mice. The animals were randomized into OCT group (100 ug/kg hypo qd for 28 d) and control group (same amount of saline solution for 28 d). 28 d later, the mice were killed to mearsure the tumor size and weight. Immunohistochemical stainning was applied to examine the expression of vascular endothelial growth factor (VEGF) and micro vessel density (MVD). Results The tumor weight and tumor size and the expression of VEGF showed remarkable differences between the two groups(P<0.05). The expression of MVD was lower in OCT group than in the control, but there was no statistic significance(P>0.05). Conclusion Octreotide could inhibit the growth of human hepatocellular carcinoma BEL-7402 cell line in nude mice. Octreotide could reduce the expression of VEGF in tumor tissue and tends to reduce the expression of MVD.
