摘    要:目的观察共轭亚油酸(CLA)对甲醛致内皮细胞株(HUVEC-12)损伤的影响及其作用机制。方法HUVEC-12体外培养至对数生长期,分别用浓度为0.0、50.0、100.0.200.0μmol/L的CLA预孵12h,再经0.1mmol/L甲醛处理,电镜观察细胞形态学变化,分光光度法检测细胞培养液中LDH、MDA和NO的含量变化。结果甲醛可致内皮细胞损伤,导致细胞膜破裂,甲醛损伤组(0.0μmoL/LCLA)细胞LDH、MDA和NO释放量高于正常对照组,差异有显著性(P〈0.01);与甲醛损伤组比较,50.0μmol/。LCLA组内皮细胞LDH释放量低(P〈0.01),100.0及200.0μmol/LCLA组LDH、MDA和NO释放量均下降(P〈0.01),但100.0及200.0μmol/LCLA组间差异无显著性。结论甲醛致内皮细胞损伤与细胞脂质过氧化有关,适当浓度的CLA可减轻这种损伤。

Abstract:Objective To study the injury and its mechanism of formaldehyde on HEVEC- 12 and the protective effects of conjugated linoleic acid (CLA). Methods HUVEC - 12 was cultured with DMEM in vitro and the cells were selected in the exponential phase of growth for experiment. The cells were pretreated with 0.0. 50.0. 100.0. and 200.0 μmol/L CLA and cultured with 0. 1mmol/L formaldehyde. The cell morphology was monitored by transmission electron microscope ( TEM). The concentrations of MDA, transudation contents of LDH and NO were measured by spectrophotography to evaluate the oxidative damage and possible mechanism. Results Formaldehyde induced cell membrane njury. The levels of LDH and MDA in 0.1 mmol/L formaldehyde group were significantly increased as compared with the control group (P〈 0.01). While preincubated CLA with cells for 12 h, LDH, MDA, and NO were reduced (P〈 0.01). Conclusions The damages on HUVEC - 12 induced by formaldehyde are involved in lipid peroxidation, and CLA can alleviate the damages.
