摘    要:采用正交试验优化重组人超氧化物歧化酶工程菌的LB培养基,得到培养基最佳配比(g/L):蛋白胨12,酵母膏5,葡萄糖1和氯化钠10.以此为基础优化基因工程菌发酵条件,确定培养基初始pH 6.5~7.5,装液量20%,菌体密度(D600) 0.55~1.4,42℃诱导表达2~3h,目的蛋白相对表达量由优化前的25%提高至4...

Abstract:The LB medium of recombinant human superoxide dismutase genenic engineering bacteria was optimized by orthogonal test.The optimal ratios of medium components(g/L) were as follows:peptone 12,yeast extract 5,glucose 1 and sodium chloride 10.The fermentation conditions were optimized in the above optimal medium.The relative expression rate of rhSOD was increased from 25%to 41%,and the absolute expression rate of rhSOD was found 2.4 times as that before optimization when the incubation parameters were adjusted ...
