人腺样囊性癌细胞系体外NT-3分泌水平的测定

摘    要:目的研究体外培养的ACC细胞系中神经营养素-3(neurotrophin-3,NT-3)的分泌情况。方法应用酶联免疫吸附测定法检测NT-3在ACC细胞系体外的分泌水平。收集ACC-M、ACC-2、M3SP4、L929 4个细胞系无血清培养的上清液,用ELISA法检测上清液中NT-3的含量。将各组细胞培养上清液中NT-3的分泌水平采用SNK-q检验进行实验组之间、实验组与对照组之间的多重比较。结果阴性对照组未检测到NT-3的表达;ACC-M、ACC-2细胞培养上清液中NT-3的分泌水平与阴性对照组比较有显著统计学意义(P〈0.01);M3SP4,及L929细胞培养上清液中NT-3的分泌水平与阴性对照组比较无显著统计学意义(P〉0.05)。ACC的两个细胞系ACC-M、ACC-2细胞分别与其他细胞系(人粘液表皮样癌高转移M3SP4细胞系、小鼠肺纤维瘤L929细胞系)NT-3的分泌水平两两比较均存在显著性差异(P〈0.01)。ACC-M细胞培养上清液中NT-3的含量与ACC-2细胞比较无显著性差异(P〉0.05)。结论体外培养的ACC细胞能分泌一定浓度的NT-3,NT-3可能在ACC的嗜神经侵袭中发挥重要作用。

Abstract:Objective To investigate the secretion of neurotrophin-3 in ACC cells in vitro. Methods Supematant of culture solution of ACC-M, ACC-2, M3SP4 and L929 were collected and NT-3 ELISA KIT was applied to measure the level of NT-3 in the supematant. SNK-q test was used to analyze the differences in NT-3 level between different groups. Results No NT-3 was detected in negative control group. There was significant difference in level of NT-3 between ACC-M/ACC-2 and negative control group (P〈0.01). There was no significant difference of NT-3 level between M3 SP4/ L929 and negative control group (P〉0.05). The level of NT-3 secreted by ACC-M/ACC-2 was significantly higher than that in M3SP4/L929 group (P〈0.01). There was no significant difference in level of NT-3 between ACC-M and ACC-2 (P〉0.05). Conclusion NT-3 can be secreted to the culture without serum by ACC cell lines and may play an important role in perineural invasion in ACC.
